【I'm with you on this.】
I agree with you. の、ややカジュアルな言い方です。on this は、「これに関して」の意味です。

【I can agree to that.】
agree with が自分も同意見であるので同意するというニュアンスなのに比べ、agree to は、提案や意見、要求などに対して、それを受け入れられる、従うことに異存はない、というニュアンスがあります。

【It IS important...】
この IS のように BE 動詞を強調することで、意味を強調することがあります。ここでは「確かに（←強調して読む）重要ですよね」と自分が強く同意することを強調しています。通常動詞を強調したい場合は、現在形であれば動詞の前に DO を、過去形ならば DID を加えます。「確かにそう思う」ならば I DO think so. となります。
